We are just now getting involved in using Amazon's S3 service as we have several big projects we are working on. Today, I set up a test product in WP eStore using the Amazon integration and it worked beautifully!

I've got my music download store working with Amazon S3 - works very well and takes the strain of large gigabyte downloads of our shared web server.
