I bought inifinity remix theme on purpose to use it on wordpress MU (v.3.0). and want to use the thumbnail picture function on the front page.
Unfortunately, the correcting the directory as explained (wp-content/uploads) is not possible on wpmu.
My files are stored like this -> http://oliviernesmon.com/files/2010/06/moutons.jpg
How can I either instruct wpmu to change the path or tweak inifinity to make it accept full url ?

Open the "index.php" file of your theme and search for the following:
timthumb.php?src=/<?php
once you find it get rid of the slash (/) so it look like the following:
timthumb.php?src=<?php
Now, you should be able to use the full URL of the image. Please note that the image must be stored on the same domain as the theme otherwise this thumbnail script doesn't work.

at last I found the time to test your suggestion.
Unfortunately It doesn't work.
Since you said that the image must be stored on the same domain as the theme,it could be the reason.
Since I'm on a wp mu with mapping domains, I really don't know what to do with that. Unless you have another suggestion, don't worry. I'll dig my way through.
